I inserted the command, CALL ROUTINE = 'defsym', just before CONDENSE_RESTRAINTS. Is it a right place for the command?

There is no 'defsym' subroutine in the standard TOP scripts, nor is it called anywhere. You can, of course, define such a routine (as you can a routine with any name) and call it from wherever you like. See examples/commands/define_symmetry.top for an example.
